The Prenylflavonoid ENDF1 Overrules Central Nervous System Growth Inhibitors and Facilitates Regeneration of DRG Neurons.

Restoration of neuronal connectivity after a lesion of the central nervous system, such as spinal cord injury, is one of the greatest challenges in modern medicine. In particular, the accumulation of factors inhibiting the growth of axons at the site of injury is a major obstacle to the improvement of structural and thus functional.

We previously investigated prenylflavonoids group derived from hops to their capacity to promote neuroregeneration. We identified a molecule called ENDF1 a very powerful tool to increase the re-growth and branching of neurites of dorsal root ganglion neurons in culture in promoting the growth substrate.

In this study, we investigated the capacity ENDF1 to promote regeneration of rat dorsal root ganglion neurons in vitro in the presence of the three main components of the extracellular matrix that acts as an inhibitor of axon growth: semaphorin 3A, Ephrin A4 and mixed with chondroitin sulfate proteoglycans. We reported that ENDF1 application significantly promoted the percentage of sensory neurons were able to regrow their neurites irrespective of their presence inhibitor, and this to an extent similar to that obtained after NGF treatment. In addition, the greatly enhanced ENDF1 total neurite length and complexity of neurites extending from neurons challenged with axon growth inhibitors.

Although the effects of NGF and ENDF1 on regeneration of neurons was similar, ENDF1 activity is not mediated by signaling through TrkA receptor, suggesting that each molecule acts via different signaling pathways. Additionally, ENDF1 not degrade cofilin phosphorylation, downstream effectors associated regeneration of RhoA / ROCK signaling pathway. Therefore, ENDF1 is pro-neuroregenerative potent factor that may help in identifying new efficient targets for regenerative therapies of the nervous system.
